چکیده

Salinization of agricultural land is an increasing problem. Because of their high tolerance to salinity, Salicornia spp. could become model species to study salt tolerance. Moreover, they also represent promising saline crops. The salinity-growth response curve for Salicornia dolichostachya Moss was evaluated at 10 different salt concentrations in a hydroponic study in a greenhouse and at 5 different seawater dilutions in an outside setting. Salt concentrations ranged between 0 mM and 500 mM NaCl (≈ seawater salinity). Plants were grown for six weeks, and morphological and physiological adaptations in different plant parts were evaluated. S. dolichostachya had its growth optimum in the range between 100 and 300 mM NaCl in the root medium, independent of the basis on which growth was expressed. The relative growth rate (RGR) in the greenhouse experiment was comparable with RGR-values in the outdoor growth-experiment. Leaf succulence and stem diameter had the highest values at the growth optimum (300 mM NaCl). Carbon isotope discrimination (δ 13 C) decreased upon salinity. S. dolichostachya maintained a lower leaf sap osmotic potential relative to the external solution over the entire salinity range, this was mainly accomplished by accumulation of Na + and Cl – . Glycine betaine concentrations did not significantly differ between the treatments. Na + :K + -ratio and K + -selectivity in the shoots increased with increasing salinity, both showed variation between expanding and expanded shoot parts. I concluded that S. dolichostachya is highly salt tolerant and showed salt requirement for

Candidate salt tolerance genes in Salicornia

To date, no gene expression comparisons have been conducted between highly salt tolerant halophytes and related glycophytes. Therefore, I investigated the effects of salinity on ion accumulation and expression of candidate salt tolerance genes in the highly tolerant salt accumulating halophyte Salicornia dolichostachya and the taxonomically related glycophytic Spinacia oleracea. S. dolichostach...

Increased vacuolar Na(+)/H(+) exchange activity in Salicornia bigelovii Torr. in response to NaCl.

Shoots of the halophyte Salicornia bigelovii are larger and more succulent when grown in highly saline environments. This increased growth and water uptake has been correlated with a large and specific cellular accumulation of sodium. In glycophytes, sensitivity to salt has been associated with an inability to remove sodium ions effectively from the cytoplasm in order to protect salt-sensitive ...

Multiple compartmentalization of sodium conferred salt tolerance in Salicornia europaea.

Euhalophyte Salicornia europaea L., one of the most salt-tolerant plant species in the world, can tolerate more than 1000 mM NaCl. To study the salt tolerance mechanism of this plant, the effects of different NaCl concentrations on plant growth, as well as Na(+) accumulation and distribution at organ, tissue, and subcellular levels, were investigated. Optimal growth and an improved photosynthet...

A SNARE-Like Superfamily Protein SbSLSP from the Halophyte Salicornia brachiata Confers Salt and Drought Tolerance by Maintaining Membrane Stability, K+/Na+ Ratio, and Antioxidant Machinery

About 1000 salt-responsive ESTs were identified from an extreme halophyte Salicornia brachiata. Among these, a novel salt-inducible gene SbSLSP (Salicornia brachiata SNARE-like superfamily protein), showed up-regulation upon salinity and dehydration stress. The presence of cis-regulatory motifs related to abiotic stress in the putative promoter region supports our finding that SbSLSP gene is in...

SOS1 expression in Salicornia

The plasma membrane Na + /H + -antiporter SOS1 is essential for salt tolerance in plants. In Chapter 4, we observed a ten-fold higher constitutive expression of SOS1 in the highly salt tolerant salt-accumulating halophyte Salicornia dolichostachya in comparison with a lower inducible SOS1 expression in its glycophytic family member Spinacia oleracea. The objective of this study was to investiga...

A Novel Gene SbSI-2 Encoding Nuclear Protein from a Halophyte Confers Abiotic Stress Tolerance in E. coli and Tobacco

Salicornia brachiata is an extreme halophyte that grows luxuriantly in coastal marshes. Previously, we have reported isolation and characterization of ESTs from Salicornia with large number of novel/unknown salt-responsive gene sequences. In this study, we have selected a novel salt-inducible gene SbSI-2 (Salicornia brachiata salt-inducible-2) for functional characterization. Bioinformatics ana...
